Abstract
The success of pharmaceutical drug delivery methods hinges on the rational design of formulations and the functional properties of their excipients. This thesis examines innovative formulation strategies to enhance drug delivery performance across both topical and non-invasive routes, with a focus on the role of excipients in regulating physical stability, therapeutic efficacy, and patient acceptability. In Chapter 1, the sustainability dimension of formulation science is explored by replacing petroleum-derived components in oil-in-water creams with greener, plant-based alternatives. The study evaluates the physical properties and functional performance of these sustainable creams, demonstrating their potential to reduce environmental burden without compromising formulation quality. Chapter 2 delves into the use of Sepineo™ P600, a novel polymeric emulsifier and gelling agent, used to develop emulgels for topical drug delivery. The relationship between rheological behavior, physical stability, and drug release performance is systematically investigated, highlighting the importance of excipient selection in achieving stable drug delivery and patient-friendly textures.
In Chapter 3, the formulation of an oral leptin antibody-based delivery system is presented as a novel approach to overcoming leptin resistance in obesity management. Challenges associated with oral delivery of biologics are addressed through strategic formulation design, targeting to protect the antibody from degradation by using specific excipients and enabling targeted leptin neutralization in the gut lumen.
Together, these studies feature the pivotal role of excipients and formulation science in advancing pharmaceutical drug delivery. By integrating sustainability, functionality, and innovation, this thesis provides a multidisciplinary perspective on designing effective and environmentally reasonable drug delivery systems.
